STAT3 but Not STAT5 Contributes to the Protective Effect of Electroacupuncture Against Myocardial Ischemia/Reperfusion Injury in Mice

چکیده
Electroacupuncture (EA) can help reduce infarct size and injury resulting from myocardial ischemia/reperfusion (I/R); however, the underlying molecular mechanism remains unknown. We previously reported that STAT5 plays a critical role in cardioprotective effect of remote ischemic preconditioning (RIPC). Here, we assessed effects electroacupuncture pretreatment (EAP) on I/R presence and/or absence Stat5 mice investigated whether EAP exerts its STAT5-dependent manner. Adult fl / Stat5- cKO were exposed to at Neiguan (PC6) for 7 days before induction by left anterior descending (LAD) coronary artery ligation. The (IS), area risk, apoptotic rate cardiomyocytes detected. RT-qPCR western blotting used measure gene protein expression, respectively, homogenized heart tissues. RNA-seq was identify candidate genes pathways. Our results showed decreased IS cardiomyocyte apoptosis. further found activated but not mice, whereas opposite observed STAT3. Following EAP, levels antiapoptotic proteins Bcl-xL, Bcl-2, p-AKT increased , while interleukin 10 (IL-10) both cKO. expression profile tissues different between with EAP. Importantly, top 30 DEGs under enriched IL-6/STAT3 signaling pathway. revealed first time protective following attributable to, dependent on, STAT5. Additionally, could activate STAT3 gene, also antiapoptotic, survival, anti-inflammatory

Heamagglutinin Conserved Domain (HA2) Prepared in Prokaryotic System is Immunogenic in Mice but not Protective against Lethal Influenza Challenge
Background and Aims: Influenza vaccine production process is time-consuming with little-to-no cross-protection which requires annual adjustment. The construction of a universal vaccine to deal with the pandemics and epidemics which occasionally threat human population is the aim of many researches worldwide. Today, influenza vaccines are mostly against two major antigenic proteins, hemagglutini...
متن کاملProtective effect of hydrogen sulphide against myocardial hypertrophy in mice
Cardiac hypertrophy is a critical component of phenotype in the failing heart. Recently, increasing evidence has demonstrated that oxidative stress plays an important role in the pathogenesis of myocardial hypertrophy. In the present study, we generated a mouse model of transverse aortic constriction (TAC) to investigate whether hydrogen sulfide (H2S) has protective effects against cardiac hype...

15 صفحه اولMediated protective effect of electroacupuncture pretreatment by miR-214 on myocardial ischemia/reperfusion injury
BACKGROUND Electroacupuncture pretreatment plays a protective role in myocardial ischemia/reperfusion (I/R) injury and microRNAs (miRNAs) could act on various facets of cardiac function. However, the role of miRNAs in the cardioprotection by electroacupuncture pretreatment on myocardial I/R injury remains unknown. The purpose of the study was to examine whether miR-214 was involved in cardiopro...
متن کاملIschemia-induced norepinephrine release, but not norepinephrine-derived free radicals, contributes to myocardial ischemia-reperfusion injury.
BACKGROUND Norepinephrine (NE)-derived free radicals may contribute to myocyte injury after ischemia -reperfusion, so the influence of sympathetic denervation on myocardial ischemia - reperfusion injury was investigated in the present study. METHODS AND RESULTS Cardiac sympathetic denervation was produced in Wistar rats by a solution of 10% phenol 1 week before ischemia. Atenolol (0.5 mg/kg) ...
